RESUMO

The aim of this study was to differentiate canine adipose-derived mesenchymal stem cells (ADMSCs) into insulin-producing cells by using culture media with different compositions to determine the most efficient media. Stem cells isolated from the fat tissues close to the bitch uterus were distributed into 6 groups: (1) Dulbecco's modified Eagle medium (DMEM)-high glucose (HG), ß-mercaptoethanol, and nicotinamide; (2) DMEM-HG, ß-mercaptoethanol, nicotinamide, and exendin-4; (3) DMEM-HG, ß-mercaptoethanol, nicotinamide, exendin-4, B27, nonessential amino acids, and l-glutamine; (4) DMEM-HG, ß-mercaptoethanol, and nicotinamide (for the initial 8-d period), and DMEM-HG, ß-mercaptoethanol, nicotinamide, exendin-4, B27, nonessential amino acids, l-glutamine, and basic fibroblast growth factor (for the remaining 8-d period); (5) DMEM-HG and fetal bovine serum; and (6) DMEM-low glucose and fetal bovine serum (standard control group). Adipose-derived mesenchymal stem cells from groups 1 to 5 gradually became round in shape and gathered in clusters. These changes differed between the groups. In group 3, the cell clusters were apparently more in numbers and gathered as bigger aggregates. Dithizone staining showed that groups 3 and 4 were similar in terms of the mean area of each aggregate stained for insulin. However, only in group 4, the number of insulin aggregates and the total area of aggregates stained were significantly bigger than in the other groups. The mRNA expression of PDX1, BETA2, MafA, and Insulin were also confirmed in all the groups. We conclude that by manipulating the composition of the culture medium it is possible to induce canine ADMSCs into insulin-producing cells, and the 2-staged protocol that was used promoted the best differentiation.

BACKGROUND: Treatment with P2Y12 receptor antagonists increases the risk for perioperative bleeding, but there is individual variation in the antiplatelet effect and time to offset of this effect. We investigated whether preoperative platelet function predicts the risk of bleeding complications in ticagrelor-treated cardiac surgery patients. METHODS: Ninety patients with ticagrelor treatment within <5 days of surgery were included in a prospective observational study. Preoperative platelet aggregation was assessed with impedance aggregometry using adenosine diphosphate (ADP), arachidonic acid (AA), and thrombin receptor-activating peptide (TRAP) as initiators. Severe bleeding complications were registered using a new universal definition of perioperative bleeding. The accuracy of aggregability tests for predicting severe bleeding was assessed using receiver operating characteristic (ROC) curves, which also identified optimal cut-off values with respect to sensitivity and specificity, based on Youden's index. RESULTS: The median time from the last ticagrelor dose to surgery was 35 (range 4-108) h. The accuracy of platelet function tests to predict severe bleeding was highest for ADP [area under the ROC curve 0.73 (95% confidence interval 0.63-0.84, P<0.001); TRAP 0.61 (0.49-0.74); AA 0.53 (0.40-0.66)]. The optimal cut-off for ADP-induced aggregation was 22 U. In subjects with ADP-induced aggregation below the cut-off value, 24/38 (61%) developed severe bleeding compared with 8/52 (14%) when aggregation was at or above the cut-off value (P<0.001). The positive and negative predictive values for this cut-off value were 63 and 85%, respectively. CONCLUSIONS: Preoperative ADP-induced platelet aggregability predicts the risk for severe bleeding complications in ticagrelor-treated cardiac surgery patients.

BACKGROUND: Wound blood is highly activated and has poor haemostatic properties. Recent data suggest that retransfusion of unwashed wound blood may impair haemostasis. We hypothesized that cell saver processing of wound blood before retransfusion reduces the negative effects. METHODS: Wound blood was collected from 16 cardiac surgery patients during cardiopulmonary bypass. One portion of the wound blood was processed in a cell saver and one portion left unprocessed. Increasing amounts of unprocessed blood (10% and 20% of the systemic blood volume) or corresponding volumes of processed blood were added ex vivo to whole blood samples from the same patient. Clot formation was assessed by modified thromboelastometry (ROTEM(®) ) and platelet function with impedance aggregometry (Multiplate(®) ). RESULTS: Addition of unprocessed wound blood significantly impaired clot formation and platelet aggregability. Cell saver processing before addition did not influence clot formation but abolished completely the negative effects of wound blood on platelet aggregability tested with all agonists. Median adenosine diphosphate-induced platelet aggregation was 51 (25th and 75th percentiles 42-69) when 20% processed cardiotomy suction blood was added vs. 34 (24-52) U when 20% unprocessed blood was added, P < 0.001. The corresponding figures for arachidonic acid-, thrombin receptor activating peptide- and collagen-induced aggregation was 21 (17-51) vs. 13 (10-25) U, 112 (87-128) vs. 78 (65-103) U and 58 (50-73) vs. 33 (28-44) U, respectively, all P < 0.001). CONCLUSION: The results suggest that cell saver processing before retransfusion mitigates the negative effects of wound blood on platelet function despite that cell saver processing reduces platelet count.

A piometra é uma afecção comum em cadelas com uma etiologia hormonal associada à infecção bacteriana, levando ao acúmulo de exsudato uterino. Desequilíbrios ácido-base e hidroeletrolíticos são complicações que contribuem para a progressão da doença, o que piora o estado geral da paciente e pode levá-la ao óbito. O objetivo do presente trabalho foi caracterizar os distúrbios ácido-base e eletrolíticos em cadelas com piometra, além de avaliar se a terapia hídrica pré-cirúrgica (Ringer lactato) é efetiva na correção desses desequilíbrios. Vinte cadelas com piometra foram submetidas à hemogasometria em oito tempos preestabelecidos. Concluiu-se que o distúrbio ácido-base mais frequente foi a alcalose respiratória e que a terapia hídrica no pré-cirúrgico com solução Ringer lactato foi efetiva na correção da acidose metabólica e proporcionou melhora na alcalose respiratória, embora não tenha corrigido quadros de alcalose metabólica.


Pyometra is a common disease in dogs with a hormonal etiology associated with a bacterial infection and leading to accumulation of uterine exudates. Acid-base and electrolyte disturbances are complications that contribute to disease progression, worsening the condition of the patient, possibly leading death. The aim of this study was to characterize the acid-base and electrolyte disturbances in dogs with pyometra, and to evaluate whether preoperative fluid therapy (Ringer's lactate) is effective in correcting these imbalances. Twenty bitches with pyometra were subjected to blood gas analysis in eight pre-set times. It was concluded that the acid-base disorder was the most frequent respiratory alkalosis and fluid therapy in the preoperative Ringer 's lactate solution was effective in the correction of metabolic acidosis, although this has not corrected metabolic alkalosis frames and has provided improved alkalosis breathing .

From fifteen female dogs with clinical diagnoses of pyometra, hematological exams were performed at three times: M0 (prior to the surgery), M24 (24h after ovarysalpingohisterectomy-OSH) and M48 (48h after OSH). Anemia was seen in 80% of the cases, characterized as mild normocytic normochromic type. The means of total leukocyte counts were 27.043, 57.940 and 40.139 céls/µL in M0, M24 and M48. A total of80% of the animals presented neutrophilic left shift in all moments. During medullar exams, the cellular, iron reserve and megakaryocytic concentration were raised as well as the ME ratio, showing a value of 26,3:1,0, probably due to the elevation of granular proliferation and maturation compartment, as the mean of the reserve compartment was within normal range. As 83% of the animals with neutrophilic left shift showed a melullary reserve compartment raised, it can be concluded that female dogs with piometra had left shift of neutrophils with disproportionally between compartments, without segmented medullar saturation.(AU)

RESUMO

The present report describes the clinical and pathological findings related to diffuse heterotopic hairs associated with pseudo-placentational endometrial hyperplasia in a canine uterus. Macroscopically, the uterus was intensely enlarged and thicker, and the endometrial surface was irregular and covered with black hairs similar to the hairs of the bitch. Histologically, heterotopic hairs associated with pseudo-placentational endometrial hyperplasia were observed.


O presente relato descreve os achados clínicos e patológicos de uma heterotopia de pelos difusa associada com hiperplasia endometrial pseudo-placentacional em um útero de uma cadela. Macroscopicamente, o útero estava intensamente aumentado de volume e espesso, a superfície do endométrio estava irregular e recoberta com pelos pretos semelhantes aos pelos da cadela. Histologicamente, heterotopia de pelos associada com hiperplasia endometrial pseudoplacentacional foi observada.

This report investigated the impact of graft-versus-host disease (GVHD) on transplantation outcomes in 1859 acute myeloid leukemia patients given allogeneic peripheral blood stem cells after reduced-intensity conditioning (RIC allo-SCT). Grade I acute GVHD was associated with a lower risk of relapse (hazards ratio (HR)=0.7, P=0.02) translating into a trend for better overall survival (OS; HR=1.3; P=0.07). Grade II acute GVHD had no net impact on OS, while grade III-IV acute GVHD was associated with a worse OS (HR=0.4, P<0.0.001) owing to high risk of nonrelapse mortality (NRM; HR=5.2, P<0.0001). In time-dependent multivariate Cox analyses, limited chronic GVHD tended to be associated with a lower risk of relapse (HR=0.72; P=0.07) translating into a better OS (HR=1.8; P<0.001), while extensive chronic GVHD was associated with a lower risk of relapse (HR=0.65; P=0.02) but also with higher NRM (HR=3.5; P<0.001) and thus had no net impact on OS. In-vivo T-cell depletion with antithymocyte globulin (ATG) or alemtuzumab was successful at preventing extensive chronic GVHD (P<0.001), but without improving OS for ATG and even with worsening OS for alemtuzumab (HR=0.65; P=0.001). These results highlight the role of the immune-mediated graft-versus-leukemia effect in the RIC allo-SCT setting, but also the need for improving the prevention and treatment of severe GVHD.

Elbow arthroscopy was performed in 30 dogs of different breeds. The procedure was performed bilaterally in 20 of these dogs, yielding a total of 50 joints. Different lesions were found, varying from cartilage fissures (8) to fragmentation (42) of medial coronoid process (FCP) of the ulna. Osteochondritis dissecans (OCD) of the humerus medial condyle was associated in four of them. All of these cases displayed varying degrees of synovitis. Osteoarthrosis (OA) in varying intensity was observed in 44 joints. The majority of cases were treated two to four months after the manifestation of clinical signs. Good clinical recovery occurred in dogs with minimal joint lesions, where these were diagnosed and treated within four weeks of the onset of clinical symptoms. Early diagnosis and arthroscopic treatment prevent osteoarthrosis and preserve locomotor function.


Artroscopia do cotovelo foi realizada em 30 cães de diferentes raças, bilateral em 20, totalizando 50 articulações. Foram encontradas diferentes lesões que variavam de fissuras na cartilagem (8) a fragmentação (42). Osteocondrose dissecante (OCD) do côndilo medial do úmero estava associada à fragmentação do processo coronoide (FCP) medial da ulna em quatro casos. Em todas articulações verificou-se sinovite em graus variados. Osteoartrose (OA) de diferentes intensidades foi observada em 44 articulações. O tratamento foi instituído na maioria dos casos, dois a quatro meses após a manifestação dos sinais clínicos. Recuperação clínica satisfatória ocorreu naqueles com mínima lesão articular, cujo diagnóstico e tratamento se fizeram dentro de quatro semanas após a manifestação clínica. O diagnóstico precoce e o tratamento artroscópico previne a osteoartrose e preserva a função locomotora.

Between 1982 and 2009 a total of 92 patients with myelofibrosis (MF) in chronic phase underwent allo-SCT in nine Nordic transplant centers. Myeloablative conditioning (MAC) was given to 40 patients, and reduced intensity conditioning (RIC) was used in 52 patients. The mean age in the two groups at transplantation was 46±12 and 55±8 years, respectively (P<0.001). When adjustment for age differences was made, the survival of the patients treated with RIC was significantly better (P=0.003). Among the RIC patients, the survival was significantly (P=0.003) better for the patients with age <60 years (a 10-year survival close to 80%) than for the older patients. The type of stem cell donor did not significantly affect the survival. No significant difference was found in TRM at 100 days between the MAC- and the RIC-treated patients. The probability of survival at 5 years was 49% for the MAC-treated patients and 59% in the RIC group (P=0.125). Patients treated with RIC experienced significantly less aGVHD compared with patients treated with MAC (P<0.001). The OS at 5 years was 70, 59 and 41% for patients with Lille score 0, 1 and 2, respectively (P=0.038, when age adjustment was made). Twenty-one percent of the patients in the RIC group were given donor lymphocyte infusion because of incomplete donor chimerism, compared with none of the MAC-treated patients (P<0.002). Nine percent of the patients needed a second transplant because of graft failure, progressive disease or transformation to AML, with no significant difference between the groups. Our conclusions are (1) allo-SCT performed with RIC gives a better survival compared with MAC. (2) age over 60 years is strongly related to a worse outcome and (3) patients with higher Lille score had a shorter survival.

RESUMO

A 7-month-old miniature poodle female dog was referred with dysphagia. After clinical, radiographic, and endoscopic examination, it was diagnosed a probable case of cricopharyngeal achalasia. The patient underwent surgical treatment and presented normal swallowing and no regurgitation after 24 hours post-surgery. Positive recovery and progressive body weight gain until 180 days after surgery was observed.


Uma cadela da raça poodle miniatura com sete meses de idade foi atendida no Hospital Veterinário apresentando quadro de disfagia. Após exames clínico, radiográfico e endoscópico detalhados, houve diagnóstico provável de acalasia cricofaríngea. A paciente, submetida a tratamento cirúrgico, apresentou deglutição normal e nenhuma regurgitação 24 horas pós-cirurgia. Houve recuperação satisfatória e ganho de peso progressivo até 180 dias após a cirurgia.

RESUMO

A case of mammary carcinosarcoma is reported in a 13-year-old, mixed breed female cat, which was not spayed and had not received contraceptives. The patient presented extensive and coalescent nodules in all mammary glands. Based on the histological and immunohistochemical findings, the diagnosis of mammary carcinosarcoma was confirmed.


Descreve-se um caso de carcinossarcoma mamário em uma gata, sem raça definida, de 13 anos de idade. A gata não era castrada e não recebeu anticoncepcionais. A paciente apresentou extensivos e coalescentes nódulos em todas as glândulas mamárias. Com base nos achados histológicos e imunoistoquímicos, firmou-se o diagnóstico de carcinossarcoma da glândula mamária.

The aim of this study was to evaluate the distribution of frozen-thawed spermatozoa within the uterine lumen and oviducts following intrauterine laparoscopic deposition at two sites. Twelve bitches of unknown reproductive history were randomly distributed into two groups. Semen (3 ml containing 300 × 10(6) frozen-thawed spermatozoa) was infused at the uterine body (UB group) or at the cranial tip of the left uterine horn. A 22-G catheter was used to access the uterine lumen. Sperm cell distribution was evaluated after ovariohysterectomy performed 3 h after artificial insemination (AI). There was no difference between groups in mean time to perform AI. Spermatozoa were detected in all uterine segments, including the tip of both horns, but none was detected in the oviduct. The 22-G catheter facilitated deposition of semen in the uterine lumen, particularly at the UB site. Sperm cell distribution occurred evenly along both horns, independent of the site of semen deposition.

Avaliaram-se e classificaram-se por meio da vídeo-artroscopia as alterações da articulação fêmoro-tíbio-patelar (FTP), 21 dias após secção experimental do ligamento cruzado cranial (LCCr) em cães, tendo como referência os parâmetros encontrados no exame artroscópico imediatamente anterior à secção. Ao exame artroscópico, foi possível visibilizar os cotos remanescentes do LCCr e as alterações articulares, como aumento da vascularização, aumento das vilosidades na membrana sinovial, bem como fibrilação e osteófitos, principalmente nas bordas trocleares do fêmur. Os resultados sugerem que a artroscopia é um método preciso de avaliação macroscópica dos tecidos articulares moles e duros.


By the use of videoarthroscopy, the changes of the stifle joint after experimental rupture of cranial cruciate ligament (CCL) in dogs were evaluated and classified, based on the parameters found at the arthroscopic examination right before the rupture. By the arthroscopic examination, it was possible to visualize the remainings of the CCL and joint changes such as increase of the vascularity and synovial proliferation increase in the synovial membranes, as well as fibrillation and irregularities on the joint surfaces and osteophyte formation, mainly on the trochlear ridges. The results suggest that arthroscopy is a precise method for macroscopic evaluation of the smooth and bone joint tissues.

As papilas mamárias de 23 búfalas foram avaliadas por meio de exame clínico e ultra-sonográfico. Em oito animais do mesmo grupo, as papilas mamárias foram submetidas ao exame teloscópico. Os resultados mostraram que o exame ultra-sonográfico e a teloscopia podem ser utilizados para a avaliação das papilas mamárias na espécie bubalina. As restrições ao uso desses exames, com a metodologia e os equipamentos empregados, foram: a identificação ultra-sonográfica dos ductos papilares foi precária para as papilas mais curtas, e em matrizes jovens, a resistência do ducto papilar limita o uso de telescopia axial.


All the mammart papillae from 23 water buffaloes were clinically and ultrasonographically examined, and eight animals from the same herd were submitted to theloscopic examination. The results showed that ultrasonography and theloscopy are satisfactory methods to evaluate the mammary papillae of the water buffalo. However, the ultrasonographic identification of the streak canal of the shorter papillae was poor and, it was not possible to proceed with axial theloscopy in some animals, specially the primiparous.

Quinze cães, sem raça definida, de ambos os sexos, de peso entre 18 e 25kg, foram submetidos à secção artroscópica do ligamento cruzado cranial (LCCr) para indução da doença articular degenerativa (DAD). Após três semanas de instabilidade articular, o LCCr foi substituído pela fáscia lata segundo a técnica de Schwalder (1989) e os animais foram distribuídos em três grupos de cinco. Os animais do grupo I, controle, não receberam tratamento medicamentoso; os do grupo II, 24mg/animal de sulfato de condroitina, por via IM, de cinco em cinco dias, totalizando seis aplicações; e os do grupo III foram tratados com hialuronato de sódio na dose de 20mg/animal, por via IV, de cinco em cinco dias num total de três administrações. Ao final de 90 dias, os animais foram eutanasiados e procedeu-se à colheita e ao processamento histológico da membrana sinovial e da cartilagem articular para avaliações morfológica e morfométrica. No grupo I foram observadas alterações degenerativas de DAD mais acentuadas que nos demais grupos, como redução do número de condrócitos, presença de pânus, fibrilações, fissuras, erosões e irregularidades na superfície articular. No grupo II observou-se elevação do número de condrócitos com aumento da atividade de síntese da matriz e redução das lesões na superfície da cartilagem. No grupo III houve aumento do número de condrócitos que eram, muitas vezes, morfologicamente inviáveis. Todos os grupos apresentaram proliferação da membrana sinovial e presença de infiltrado linfoplasmocitário na subíntima e na perivascular. Nos grupos I e III, a proliferação da membrana sinovial era exuberante com formação de pânus, presença de sinoviócitos achatados ou ausência de sinóvia com tecido de granulação. Os resultados sugerem que o sulfato de condroitina estimulou a cartilagem articular, diminuindo ou retardando as alterações da DAD e o hialuronato de sódio não interferiu no processo degenerativo da cartilagem articular. Fifteen mongrel dogs, both genders, weighting from 18 to 25kg were used and Degenerative Joint Disease (DJD) was induced through cranial cruciate ligament (CCrL) artroscopical section. After three weeks, CCrL was reconstructed by Schawalder's (1989) technique. Then, dogs were distributed in three groups and the following protocols were used: group I, control, no other treatment but the CCrL reconstruction; group II received chondroitin sulfate 24mg per animal every five days, intramuscularly, in a total of six injections; and group III received sodium hyaluronate 20mg per animal every five days, intravenously, in a total of three injections. Clinical observation was done until 90 days after treatments. By that time, the articular cartilage and synovium were collected and their morphology was evaluated. In group I, the degenerative alterations of the DJD were the most intense. Thus, decrease of chondrocytes number, pannus, fibrillations, grooves, erosion, and irregular articular surface were observed on the cartilage. In group II, raise of chondrocytes number was observed, with increase of synthesis activity of matrix and decrease of lesions on the articular surface. There was an increase of chondrocytes in group III, but the cells were morphologically unviable. All the groups showed proliferation of the synovial membrane, with limpho-plasma cells infiltrated in subintim and perivascular. In groups I and III, the proliferation of synovium was abundant, with formation of pannus, flattened synoviocytes or synovium absent with granulation tissue. Those results suggest that the chondroitin sulfate stimulated the articular cartilage; decreasing or delaying the alterations of DJD, as well as, the sodium hyaluronate did not interfere on degenerative process in articular cartilage. No favorable action of these drugs in the synovial membrane was verified.

In a previous study, we found a high (33%) human papillomavirus (HPV) DNA prevalence among first year university students in the Helsinki metropolitan area. We have now performed HPV rescreening among first-round HPV-positive students using a liquid-based hybridization assay. A total of 212 students participated in rescreening, and 82 (38.7%) of 212 were found to be positive for HPV DNA. Low-risk (lr) HPV DNA was repeatedly found in 16.8% of the patients who had been lr positive in the first screening round. High-risk (hr) HPV DNA was repeatedly found in 33.3% of the patients. Although HPV typing in these samples has not been carried out yet, we conclude that repeatedly positive HPV DNA findings were strikingly common. hrHPV DNA was repeatedly found twice as often as lrHPV DNA. HPV DNA prevalence was higher among oral contraceptive users than among patients using other contraception.

Trinta e duas papilas mamárias de vacas da raça Holandesa, em período seco, foram submetidas a telotomia lateral que produziu defeito linear na mucosa da parte papilar do seio lactífero (PPSL). Houve excisão de um retângulo de mucosa de tamanho padronizado que provocou um defeito retangular na mucosa do PPSL, oposto à telotomia. Todas as telotomias foram suturadas e, aleatoriamente, em 16 delas foram introduzidas sondas de Foley de 2,7mm de diâmetro, formando o grupo de papilas com dilatador. A distensão dos balonetes das sondas de Foley provocou a dilatação da PPSL o que forçou a manutenção das sondas, por sete dias, na papila mamária. As 16 papilas restantes formaram o grupo de papilas sem dilatador. Foram realizadas videoteloscopias antes (dia 0) e após as telotomias (dia 8, após a retirada das sondas de Foley e dia 15). As avaliações morfológica e histológica do processo de cicatrização dos defeitos lineares e retangulares evidenciaram que o uso de dilatador na PPSL auxiliou na orientação cicatricial, mantendo a patência do seio lactífero em um maior número de papilas, quando os dois grupos foram comparados. A dilatação da PPSL interferiu na cicatrização das telotomias, e provocou maior número de alterações no epitélio de revestimento do seio lactífero


Thirty two mammary teats of Holstein cows, in the dry period, were submitted to histological examination. Lateral thelostomy was performed in all teats, producing a linear defect on the mucous membrane of the lactiferous sinus (MMLS). The teats were also submitted to a standard rectangular excision on the mucous membrane of MMLS, on the opposite side of thelostomy. All thelostomies were sutured and then the teats were divided in 2 groups. One group received Foleys's catheter of 2.7mm in diameter and was designed teat distention group. The 16 remain teats formed the no teat distention group. The Foleys's catheter balloon distention produced MMLS dilatation and also a 7 days maintenance period on the mammary teat. The scar evaluation of the linear and rectangular defects, performed trough morphologic and histological examinations showed that the dilatation on MMLS contributes on the scar orientation, keeping the lactiferous sinus patency in a greater number of teats, when both groups were compared. The MMLS dilatation interferes with telothomies healing, can observe so more alterations on the mucous membrane of lactiferous sinus

Avaliaram-se os aspectos clínicos e radiográficos do enxerto de pericárdio bovino, preservado em glicerina, como substituto do ligamento cruzado cranial. Quinze cães machos, sem raça definida, pesando entre 17,4 e 31,6 kg, foram submetidos à ruptura experimental do ligamento cruzado cranial e à substituição por pericárdio, via videoartroscopia. Os cães foram divididos em três grupos de cinco e avaliados aos 30, 90 e 120 dias. O membro operado foi imobilizado por duas semanas e procederam-se avaliações clínicas semanais. Radiografias foram feitas mensalmente e foram realizadas colheita de líquido sinovial nos tempos descritos. Clinicamente, os cães mostraram claudicação acentuada a moderada, hipotrofia muscular no membro operado e acentuado deslocamento cranial da tíbia em relação ao fêmur. As alterações degenerativas foram observadas nas radiografias. Observou-se instabilidade acentuada em todos os animais. O líquido sinovial tinha características de inflamação. Concluiu-se que o enxerto rompeu precocemente, provocou reação inflamatória persistente e fenômenos de rejeição, não sendo, portanto, recomendado para substituição do ligamento cruzado cranial de cães


The clinical and radiographic aspects of the bovine pericardium preserved in glicerin, were evaluated as a substitute for canine cranial cruciate ligament. Fifteen male mongrel dogs weighing between 17.4 and 31.6kg had the ligament experimentally ruptured and the stifle joint stabilized by an arthroscopical technique with bovine pericardium as a graft. The dogs were divided into three groups of five animals each. They were evaluated at 30, 90 and 120 days. The operated limb was imobillized for two weeks and clinical examination was performed weekly. Radiographs were taken monthly and the sinovial fluid was collect at 30, 90 and 120 days. Clinically, dogs presented high to moderate lameness, muscle hipotrophy in the operated limb and accentuated cranial drawer movement. Degenerative disease was detected in radiography. All dogs showed total rupture of the graft. Sinovial fluid analysis showed characteristics of inflammation. It can be concluded that pericardium graft failed prematurely, incited persistent inflammatory reaction and rejection phenomena. Thus, it cannot be recommended as a xenograft for cranial cruciate ligament replacement

OBJECTIVE: The authors evaluated the accuracy of the fine needle aspiration cytology technique in the diagnosis of 77 canine mammary gland tumours using the same cytological and histological criteria currently applied to the diagnosis of human breast cancer. METHODS: The study was performed in 73 pure or mixed-breed female dogs submitted to surgical resections of 'mammary tumours'. All cytological smears were stained by routine May-Grunwald-Giemsa and Papanicolaou stains. RESULTS: We obtained a correct cyto-histological correlation in 52/77 cases (67.5%) when all cytopathological examinations were considered, and in 52/56 cases (92.9%) when the inconclusive cases were excluded from the analysis. CONCLUSION: Our results demonstrate that, because of the similarity of the cytological findings in the human and canine mammary gland tumours, it is possible to use the same cytological criteria applied in human pathology for the diagnosis of canine mammary gland tumours.

Human papillomavirus (HPV) causes cervical, vulvar, and vaginal cancers, precancerous dysplasia, and genital warts. We report data for the longest efficacy evaluation to date of a prophylactic HPV vaccine. In total, 552 women (16-23 years) were enrolled in a randomised, placebo-controlled study of a quadrivalent HPV 6/11/16/18 L1 virus-like-particle vaccine with vaccination at months 0, 2, and 6. At regular intervals through 3 years, subjects underwent gynaecologic examination, cervicovaginal sampling for HPV DNA, serum anti-HPV testing, and Pap testing, with follow-up biopsy as indicated. A subset of 241 subjects underwent two further years of follow-up. At 5 years post enrollment, the combined incidence of HPV 6/11/16/18-related persistent infection or disease was reduced in vaccine-recipients by 96% (two cases vaccine versus 46 placebo). There were no cases of HPV 6/11/16/18-related precancerous cervical dysplasia or genital warts in vaccine recipients, and six cases in placebo recipients (efficacy = 100%; 95% CI:12-100%). Through 5 years, vaccine-induced anti-HPV geometric mean titres remained at or above those following natural infection. In conclusion, a prophylactic quadrivalent HPV vaccine was effective through 5 years for prevention of persistent infection and disease caused by HPV 6/11/16/18. This duration supports vaccination of adolescents and young adults, which is expected to greatly reduce the burden of cervical and genital cancers, precancerous dysplasia, and genital warts.
